    2-Arachidonoyl glycerol (2-AG) has been isolated from porcine brain, and has been characterized as the natural endocannabinoid ligand for the CB1 receptor. The congener of 2-AG in which a linoleoyl group replaces the arachidonoyl group is 2-linoleoyl glycerol (2-LG), and this compound also appears in vivo in conjunction with 2-AG. Although the intrinsic activity of 2-LG is low, it potentiates the activity of other endocannabinoids, including 2-AG. This “entourage” effect is attributed to blockade of the reuptake and breakdown pathways that normally function to reduce endocannabinoid levels rapidly upon release.

    Arachidonyl ethanolamide (AEA) is an endogenous lipid neurotransmitter with cannabinergic activity. It binds to both the central cannabinoid (CB1) and peripheral cannabinoid (CB2) receptors. Fatty acid amide hydrolase (FAAH) is the enzyme responsible for hydrolysis and inactivation of AEA. Metabolism of AEA by cyclooxygenase-2, leading to the formation of prostaglandin ethanolamides, and by lipoxygenases been documented as well. (±)5(6)-EET ethanolamide is a cytochrome P450 (CYP450) metabolite of AEA, although specific stereochemistry rather than a racemic mixture ensues from enzymatic metabolism, it is a potent and selective agonist of CB2 (Kι = 8.9 nM for CB2, 11.4 μM for CB1), effectively suppressing forskolin-stimulated accumulation of cAMP through this Gi-coupled receptor (IC50 = 9.8 nM).

    Lipoxins are trihydroxy fatty acids containing a 7,9,11,13-conjugated tetraene. Lipoxin A4 (LXA4) was first described as a metabolite of 15-HpETE and/or 15-HETE when added in vitro to isolated human leukocytes. Material obtained in this manner consists of four distinct isomers: 5(S), 6(S); 5(S), 6(R); and the 11-trans and 11-cis isomers of each of these. 5(S),6(S)-LXA4 is an original four metabolite that was first identified by Serhan, Nicolaou, and Samuelsson. It was considered to be an artifact by these authors because it lacked the potency of the 5(S),6(R) isomer with respect to the contraction of isolated guinea pig lung parenchymal strips. It has not been possible to isolate “natural” LXA4 from humans or other mammals in amounts sufficient for determination of absolute stereochemistry. Most authors refer to LXA4 as the 5(S),6(R), 11-cis isomer, but it is not clear that biological systems are aware of or agree with these conventions. In the past conjugated tetraenes flanked by hydroxyl groups of mixed stereochemistry have been marketed as 'Lipoxin A4' by some biomolecular supply companies. Availability of single pure LXA4 enantiomers should help to reduce the confusion this has caused.

In angiotensin-infused rats, a dosage of 25 mg/l AUDA administered in drinking water decreased mean arterial blood pressure from 161 ± 4 mmHg to 140 ± 5 mmHg. This hypotensive effect was accompanied by an elevation of urinary epoxide-to-diol ratios. AUDA activates peroxisome proliferator-activated receptor α (PPARα) 3-fold at a concentration of 10 µM and exhibits no affect on PPARδ or PPARγ.
